核因子κB受体活化因子配体(RANKL),亦称为肿瘤坏死因子配体超家族成员11(TNFSF11)、TNF相关活化诱导细胞因子(TRANCE)、骨保护素配体(OPGL)及破骨细胞分化因子(ODF),是一种II型跨膜蛋白,属于肿瘤坏死因子(TNF)超家族成员。
RANKL通过刺激破骨细胞形成与活化,成为骨吸收及整体骨密度调控的关键介质。研究还表明某些癌细胞(特别是前列腺癌细胞)可激活骨重塑进程,最终促进骨形成[17]。这种骨重塑与骨形成的增强会加速骨转移灶的生长。骨重塑的整体调控取决于RANKL与其受体RANK及诱骗受体骨保护素(OPG)的特异性结合。
