信号转导与转录激活因子3（STAT3）调控着对细胞生存、增殖、自我更新、血管生成及免疫应答等关键生物学过程至关重要的基因表达。作为早期肿瘤诊断标志物，研究认为模型系统中STAT3的持续性激活是诱发恶性转化与肿瘤发展的充分因素。在癌细胞中经典的IL-6/STAT3信号通路中，IL-6与膜结合型IL-6受体α（IL-6R）及IL-6受体β（即gp130）结合形成复合物，激活JAKs的磷酸化，进而引发STAT3磷酸化并活化。随后，磷酸化STAT3形成同源二聚体转移至细胞核内，结合靶基因启动子区域并激活其转录过程。
