Abiprubart (KPL-404) is a humanized monoclonal antibody of the IgG4 isotype, engineered with a stabilized and functionally-silent Fc region. This design is critical as it allows the antibody to bind to its target and inhibit signaling without triggering effector functions, such as antibody-dependent cellular cytotoxicity (ADCC) or complement-dependent cytotoxicity (CDC), thereby avoiding the depletion of target cells (lymphocytes/B cells). It is currently under clinical investigation for the treatment of autoimmune diseases, including rheumatoid arthritis and Sjögren's Syndrome.
