Manfidokimab (also known as AK120) is a humanized IgG4 monoclonal antibody that specifically targets the interleukin-4 receptor alpha subunit (IL‑4Rα). By binding to IL‑4Rα, it blocks the signaling of both interleukin‑4 (IL‑4) and interleukin‑13 (IL‑13), two key cytokines that drive type 2 inflammatory responses. This dual inhibition suppresses the downstream pathways involved in IgE production, eosinophil activation, mucus hypersecretion, and epithelial barrier dysfunction. As an IgG4 isotype, it is designed to minimize effector functions. It is being developed for the treatment of moderate‑to‑severe atopic dermatitis and other type 2 inflammatory diseases such as asthma.
