研究背景：SOCS-1 是 I 型和 II 型干扰素 (IFN) 信号通路以及其他细胞因子 (包括 IL2、IL4、IL6 和白血病抑制因子 (LIF)) 的关键负调控因子。它通过抑制 JAK/STAT 信号通路来下调细胞因子信号传导，具体机制是与 JAK 蛋白和 IFNGR1 结合并抑制其激酶活性。在体外实验中，SOCS-1 还能抑制 Tec 蛋白酪氨酸活性。此外，它可能作为 ECS (Elongin BC-CUL2/5-SOCS-box 蛋白) E3 泛素连接酶复合物的底物识别组分，介导靶蛋白的泛素化和随后的蛋白酶体降解。根据相似性研究，SOCS-1 还参与调节 IFN-gamma (IFNG) 介导的感觉神经元存活。

The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
