研究背景：Tau 是一种促进微管 microtubule 组装和稳定性的蛋白质，可能参与神经元极性 neuronal polarity 的建立和维持。其 C 端结合轴突微管 axonal microtubules，而 N 端结合神经质膜成分 neural plasma membrane components，表明 Tau 作为连接蛋白 linker protein 在两者间发挥作用。TAU/MAPT 在神经元细胞中由中心体 centrosome 定义的细胞体域定位，决定了轴突极性 axonal polarity 的预设。较短的 Tau 异构体 short isoforms 赋予细胞骨架 cytoskeleton 可塑性，而较长的异构体 longer isoforms 可能更倾向于稳定细胞骨架。类似功能还包括：微管结合与神经元形态调控（by similar）。

The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
