Axatilimab is a high-affinity, humanized IgG4 monoclonal antibody designed as an antagonist of the colony-stimulating factor 1 receptor (CSF-1R; also known as c-FMS). It functions by selectively binding to CSF-1R, blocking the interaction with its ligands CSF-1 and IL-34. This blockade inhibits the survival, proliferation, and differentiation of monocytes and macrophages dependent on CSF-1R signaling. It is in clinical development for the treatment of chronic graft-versus-host disease (cGVHD) and other fibrotic or inflammatory conditions, based on the rationale of depleting or reprogramming pathogenic macrophages that drive tissue inflammation and fibrosis.
