FOXO4 is a key molecule that promotes the survival of senescent cells, consisting of a 44-amino-acid polypeptide. FOXO4 can interact with the protein p53, a well-known senescence-inducing protein. Researchers inferred that this FOXO4-p53 interaction is crucial for the survival of senescent cells, and disrupting this interaction would induce apoptosis. To test this, they synthesized a modified peptide fragment called FOXO4-D reversal (FOXO4-DRI). This peptide fragment lacks the normal transcriptional activity of FOXO4 but binds more stably to p53, thereby competitively inhibiting the FOXO4-p53 interaction.
