H-Glu-Ala-pNA, a chromogenic substrate peptide for broad-spectrum aminopeptidases and metalloproteinases, is a small-molecule peptide composed of two amino acids and a fluorescent modification. Upon enzymatic cleavage, H-Glu-Ala-pNA releases p-nitroaniline (pNA, λmax 405 nm), enabling continuous spectrophotometric determination of enzyme activity. It is employed to investigate the specific recognition mechanisms of proteases for N-terminal Glu or distinct peptide bond positions. In high-temperature peptide synthesis systems, H-Glu-Ala-pNA serves as a model substrate to characterize the activity of recombinant pseudomurein endoisopeptidase (PeiW/PeiP), addressing issues such as instability and time-consuming processes associated with traditional natural substrates.
