Fumaric acid (CAS 110-17-8), also known as allomaleic acid, is a naturally occurring bioactive compound primarily extracted from plant sources such as Fumaria officinalis , lichens, mosses, and mushrooms. It is also found in animal tissues (e.g., fresh beef). As an unsaturated organic acid, it serves as an essential intermediate in the tricarboxylic acid (TCA) cycle. Due to its hydrophilic structure, fumaric acid is stable and dispersible in aqueous environments; it is generally recognized as safe (GRAS) as a food-grade acidulant and a low-molecular-weight organic acid.

The benefits of fumaric acid stem largely from its potent antimicrobial activity. Its minimum inhibitory concentration (MIC) values against Candida tropicalis , Candida krusei, Candida albicans, Candida glabrata, and Candida parapsilosis are 9,375, 37.5, 4,687, 4,687, and 1,172 mg/mL, respectively. Research indicates that bacterial cells are approximately one hundred times more sensitive to fumaric acid than Candida cells. The MIC values are determined to be 150 μg/mL for Klebsiella pneumoniae, Escherichia coli, Pseudomonas aeruginosa, and Enterobacter aerogenes, and 75 μg/mL for Staphylococcus aureus. Notably, the inhibitory concentrations for bacteria (75–150 μg/mL) are significantly lower than those for fungi (1.172–37.5 mg/mL), demonstrating high selective toxicity against bacteria. Consequently, when used as a food additive, fumaric acid not only provides high acidity—delivering a unique, intense, and lingering sour taste—but also helps extend the shelf life of food products [1].
Furthermore, it exhibits potential analgesic, anti-inflammatory, antioxidant, anti-psoriatic, chemopreventive, immunomodulatory, and neuroprotective properties.
Fumaric acid and its esters possess free-radical scavenging properties, making them excellent antioxidants; furthermore, they offer pharmacological benefits due to their anti-inflammatory, analgesic, immunomodulatory, and chemopreventive properties.
Dimethyl fumarate (CAS 624-49-7), abbreviated as DMF, is an α,β-unsaturated ester of fumaric acid that exhibits neuroprotective effects. On one hand, it exerts protective effects in neurons and oligodendrocytes by activating antioxidant defenses, enhancing respiratory function, and inhibiting apoptosis. On the other hand, it suppresses the overactivation of immune cells (such as T cells) by impairing their mitochondrial function—a mechanism consistent with its immunomodulatory benefits in the treatment of multiple sclerosis. DMF is widely used clinically to treat relapsing multiple sclerosis and plaque psoriasis [2].
