比哌立登性质、用途与生产工艺
Biperiden(KL 373)有抗帕金森症活性,是中枢M1胆碱受体阻断剂。
化学性质
结晶体。熔点112-116℃(101℃)。不易溶于水,微溶于乙醇,易溶于甲醇。该品的盐酸盐(C21H29NO·HCI)又称安克痉(Akineton,Akinophyl),为白色结晶。约275℃分解,也有报道其熔点为238℃。
用途
该品为抗震颤麻痹药、解痉药。小鼠口服LD50为545mg/kg,静脉注射为56mg/kg。
用途
Biperiden is antiparkinsonian; non-selective muscarinic receptor antagonist. Biperiden is used for the adjunctive treatment of all forms of Parkinson鈥瞫 Disease (postencephalitic, idiopathic, and arter
iosclerotic); also commonly used to improve parkinsonian signs and symptoms related to antipsychotic drug therapy.
生产方法
以环戊二烯为原料,先与氯乙烯合成氯代双环庚烯,再与镁和乙醚反应,得双环庚烯镁化氯,最后与β-哌啶苯丙酮缩合制得比哌立登。β-哌啶苯丙酮的制备:以苯乙酮为原料与哌啶盐酸盐、甲醛、二甲胺经曼氏反应得到。
