Synthesis of Compound 10

Synthesis of Compound 10 Fuming sulfuric acid (500 g, 5.1 mol) was added slowly to 2,4-lutidine 8 (55 mL, 0.48 mol) and cooled in an ice bath with stirring. Potassium nitrate (87.5 g, 0.86 mol) was then added slowly. The reaction mixture was stirred at room temperature for 1 h and then allowed to warm to 110° C. for additional 7 h, cooled to room temperature and stirred overnight. The reaction mixture was poured onto ice (1.0 kg) and neutralized to pH 9 with solid NaOH, extracted with ether. The combined extracts were dried over anhydrous Na2 SO4. The solvent was removed and the residue was distilled under reduced pressure to give 37.83 g (53%) of a mixture of 3-nitrolutidine 10 and 5-nitrolutidine 9. Further separation by distillation provided pure 3-nitrolutidine 10. 1 H NMR (CDCl3) δ 2.35 (s, 3H), 2.57 (s, 3H), 7.13 (d, 1 H, J=4.8 Hz), 8.46 (d, 1 H, J=5.0 Hz).
