One equivalent of N-alpha-fluorenemethyloxycarbonyl-N-epsilon-tert-butyloxycarbonyl-L-lysine was dissolved in dichloromethane. One part (w/w) of a 15% trifluoroacetic acid solution in dichloromethane was added. The mixture was stirred vigorously and cooled externally with tap water. Thin-layer chromatography confirmed the reaction was complete. The solvent and trifluoroacetic acid were removed by vacuum distillation. The mixture was dissolved again in dichloromethane and distilled again under vacuum. The solid was washed with petroleum ether and discarded. The remaining solid was dissolved in dimethylformamide. Two equivalents of N-methylmorpholine were added, and the pH was adjusted to 8. 0.7 equivalents of N-hydroxysuccinimide biotin were slowly added. The reaction was allowed to proceed for 3 hours. Thin-layer chromatography showed the reaction was complete. Citric acid solution was added to the reaction mixture, and the pH was adjusted to 3. The solid precipitated, was washed, and dried to obtain the product with a purity of 99.1% and a yield of 90%. |
