The general procedure for the synthesis of 3-bromo-2-methoxyaniline from 2-bromo-6-nitroanisole was as follows: 1-bromo-2-methoxy-3-nitrobenzene (20.1 g, 86.6 mmol) was heated to 80 °C with iron powder (33.4 g, 598 mmol) in a solvent mixture of acetic acid/water (1:1, 600 mL). After the reaction lasted for 1.5 h, the mixture was cooled to room temperature and filtered. The filtered solid was washed several times with ethyl acetate and water. The organic layers were combined, washed with saturated aqueous NaHCO3, dried over anhydrous sodium sulfate and concentrated under reduced pressure. The residue was purified by column chromatography (eluent: petroleum ether/ethyl acetate = 20:1 to 10:1) to afford 3-bromo-2-methoxyaniline as a yellow solid (17.4 g, 99% yield).1H-NMR (300 MHz, DMSO-d6) δ (ppm): 3.66 (s, 3H), 5.23 (s, 2H), 6.64-6.76 (m, 1H). 3H). | [References]
