The general procedure for the synthesis of 2-(1H-indol-1-yl)ethanol from indole and 2-bromoethanol is as follows: the preparation of methyl 2-[4-[2-indolyl]ethoxy]phenylthio-butanoate (ST1983) is described in Example 8. First, the intermediate 1-(2-hydroxyethyl)indole was prepared in a procedure that referenced the method reported in J. Med. Chem. 1998,41/10,1619-1639, but with the reaction time adjusted to 30 h (30 min in the original literature). The procedure was as follows: indole (5.0 g, 42.7 mmol) was dissolved in 50 ml of anhydrous DMSO, KOH (3.6 g, 64.1 mmol) and 2-bromoethanol (6.4 g, 51.3 mmol) were added and the reaction was carried out at 25-30 °C. Finally, 5 g of oily product was obtained in 73% yield.
