 1.Dissolve 5'-Dimethoxytrityl-N 4-benzoyl-2',2'-difluoro-2'-deoxycytidine (1 equiv., 0.4 mmol, 270 mg)in anhydrous acetonitrile (6 ml).
2.Add 0.5 M solution of ethylothiotetrazole (1.6 equiv., 0.65 mM, 1.3 ml) in anhydrous acetonitrile and 2-cyanoethyl-bis-(N,N'-diisopropropyl)phosphoramidite (1.43 equiv., 0.57 mmol, 0.2 ml) to the above solution.
3.Stir the reaction mixture at room temperature for 1 h to obtain product. |
