To a methanolic (1 L) suspension of 5-bromo-2-chloropyrimidin-4-amine (137.3 g, 658 mmol) was added sodium methanolate (83.5 g, 1.54 mol) in batches. The reaction mixture was stirred under reflux conditions for 2 hours. Upon completion of the reaction, the mixture was concentrated to about 400 mL and poured into saturated aqueous ammonium chloride solution (1.2 L). After stirring for 15 minutes, the aqueous layer was extracted with ethyl acetate. The combined organic layers were washed with brine, dried over anhydrous sodium sulfate, filtered and concentrated to give 5-bromo-2-methoxypyrimidin-4-amine (133.7 g, 99.4% yield). | [References]
