1. Acetone aldehyde dimethyl acetal (10 g, 84 mmol) and N,N-dimethylformamide dimethyl acetal (10 g, 84 mmol) were dissolved in DMF (30 ml) in a round-bottomed flask and the reaction was heated for 15 h at 110 °C.
2. an aqueous solution of guanidine hydrochloride (12 g, 127 mmol) and NaOH (6.7 g, 169 mmol) was added to the reaction mixture.
3. The mixture was refluxed for 36 hours.
4. After completion of the reaction, the reaction material was cooled and filtered.
5. The product was purified by recrystallization from hot ethyl acetate to afford 2-amino-4-dimethoxymethylpyrimidine (10 g, 70% yield) as a white crystalline solid. | [References]
