The general procedure for the synthesis of 6-(methylsulfonyl)-3-pyridinamine from 2-(methylsulfonyl)-5-nitropyridine was as follows: 2-(methylsulfonyl)-5-nitropyridine (1.80 g, 8.90 mmol) was dissolved in a mixed solvent of water (25 mL) and methanol (25 mL). Iron powder (1.49 g, 26.68 mmol) and ammonium chloride (2.86 g, 53.47 mmol) were added. The reaction mixture was heated to reflux temperature and stirred for 1 hour. After completion of the reaction, the mixture was filtered and the solid was washed with ethyl acetate (EtOAc). The filtrate was extracted with ethyl acetate and the organic phases were combined and dried with anhydrous magnesium sulfate (MgSO4). The desiccant was removed by filtration and the filtrate was collected and concentrated under reduced pressure to afford 6-(methylsulfonyl)-3-pyridinamine (1.40 g, 91% yield). The product was analyzed by LC-ES/MS, m/z 173 [M + H]+. | [References]
