The commercial production of amisulbrom follows a concise three-step sulfonamide coupling process. First, 3-bromo-6-fluoro-2-methylindole is N-sulfonylated with 1H-1,2,4-triazole-1-sulfonyl chloride. The resulting indolyl sulfonyl triazole is then reacted with chlorosulfonyl isocyanate in dichloromethane, followed by in situ dimethylamine addition to form the sulfonamide linkage.
