1-Acetyl-4-methylpiperazine Hydrochloride is used in the preparation of (acetyl)piperazinium iodide and pyrrolopyrazinium iodides as nicotinic agonists. | [Synthesis]
The general procedure for the synthesis of 1-acetyl-4-methylpiperazine from N-methylpiperazine and acetic anhydride was as follows: N-methylpiperazine (2.0 g, 19.97 mmol), triethylamine (3.35 mL, 23.96 mmol) and acetic anhydride (2.3 mL, 23.96 mmol) were dissolved in ethanol (60 mL) and the reaction was stirred at room temperature overnight. After completion of the reaction, the reaction mixture was concentrated to dryness under reduced pressure. The residue was purified by silica gel column chromatography using a mixed solvent of dichloromethane and methanol (with a gradient increase in the proportion of methanol) as eluent to afford the target product 1-acetyl-4-methylpiperazine (2.16 g, 76% yield) as a yellow oil. The product was characterized by 1H-NMR (CDCl3, 300 MHz) with chemical shifts δ: 3.65 (t, J = 6.8 Hz, 2H), 3.49 (t, J = 6.8 Hz, 2H), 2.46-2.39 (m, 4H), 2.32 (s, 3H), 2.10 (s, 3H) ppm. | [References]
