4-Amino-4'-chlorodiphenyl sulfone is a substituted 4-aminodiphenylsulfone as Escherichia coli dihydropteroate synthase inhibitor. | [Synthesis]
Step 2. N-[4-(4-Chlorophenyl)sulfonylphenyl]acetamide (5.5 g, 0.014 mol) was dissolved in a mixture of concentrated hydrochloric acid (70 mL) and ethanol (70 mL), and the reaction was refluxed overnight. The progress of the reaction was monitored by thin layer chromatography (unfolding reagent: petroleum ether/ethyl acetate = 2:1, Rf = 0.3) to confirm the completion of the reaction. The reaction mixture was concentrated under reduced pressure and the residue was neutralized with saturated sodium bicarbonate solution to pH>7 and subsequently extracted with ethyl acetate (80 mL x 3). The organic phases were combined, dried over anhydrous sodium sulfate and concentrated under reduced pressure to afford 4-((4-chlorophenyl)sulfonyl)aniline (4.1 g, 87% yield) as a light brown solid. | [References]
