Step C: Preparation of 3-hydroxyisonicotinonitrile: 3-(2-(trimethylmethylsilyl)ethoxy)isonicotinonitrile (1.30 g, 5.90 mmol) and anhydrous THF (5 mL) were added to a dry reaction flask, followed by slow addition of ammonium fluoride (11.8 mL, 11.8 mmol) to the THF solution. The reaction mixture was stirred at room temperature for 1 h, after which the solvent was removed by concentration via rotary evaporator. The crude product was purified by silica gel column chromatography using sequentially dichloromethane and dichloromethane solution containing 5% methanol as eluents to afford the target product 3-hydroxyisonicotinonitrile (0.7 g, 99% yield) as a dark brown oil. | [References]
