[Synthesis]
General procedure for the synthesis of 6-bromo-N-methylquinolin-4-amine from 6-bromo-4-chloroquinoline and methylamine: 6-bromo-4-chloroquinoline (100 mg, 0.41 mmol) was mixed with methylamine (2.0 M in methanol, 2 mL, 4 mmol) and placed in a Biotage Initiator microwave synthesizer and reacted for 3 hr at 120 °C. Upon completion of the reaction, the mixture was cooled, concentrated under reduced pressure and purified by fast column chromatography (eluent: dichloromethane/methanol gradient, 0-10% methanol) to afford 6-bromo-N-methylquinolin-4-amine as a white solid (40 mg, 41% yield).1H NMR (400 MHz, chloroform-d) δ ppm: 8.39 (d, J = 5.81 Hz, 1H) , 8.04 (d, J = 1.77 Hz, 1H), 7.80 (d, J = 9.09 Hz, 1H), 7.67 (dd, J = 8.84, 2.02 Hz, 1H), 6.37 (d, J = 5.81 Hz, 1H), 3.02 (s, 3H). |
