以2,4-二氯-7H-吡咯并[2,3-d]嘧啶为起始原料合成2,4-二氯-5-碘-7H-吡咯并[2,3-d]嘧啶的一般步骤:向50L夹套反应器中加入2,4-二氯-7H-吡咯并[2,3-d]嘧啶(950g,5.053mol)和二氯甲烷(16L)。将得到的棕褐色悬浮液冷却至16℃,并在20分钟内分批加入N-碘代琥珀酰亚胺(1598g,7.104mol)。将反应混合物在室温下搅拌16小时,随后通过薄层色谱(TLC)分析(展开剂比例为2:1的己烷/乙酸乙酯)确认反应完全。过滤所得沉淀,用二氯甲烷(3×1.5L)洗涤,并在40℃下减压干燥64小时,得到1447g(产率:91%)的2,4-二氯-5-碘-7H-吡咯并[2,3-d]嘧啶,为米色固体。质谱(ESI)m/z 314.0 [M + 1]+。
