在0℃及氮气保护下,向搅拌的1-(2,4-二甲基苯基)-2-[(3-甲氧基苯基)亚甲基]酰肼(500mg,1.97mmol)和三乙胺(0.33mL,2.36mmol)的二氯甲烷(5mL)溶液中缓慢滴加三氟乙酸酐(0.33mL,2.36mmol)。反应混合物在0℃下持续搅拌1.5小时。反应完成后,将混合物浓缩,残余物通过柱色谱法使用丙酮/己烷(1:10)作为洗脱剂进行纯化,得到目标产物2,2,2-三氟乙酸1-(2,4-二甲基苯基)-2-[(3-甲氧基苯基)亚甲基]酰肼(571mg,收率83%),为浅黄色油状物。1H NMR(CDCl3)数据如下:δ7.26-7.23(m,4H),7.19(d,J = 8.0Hz,1H),7.12(d,J = 7.5Hz,1H),7.03(d,J = 8.0Hz,1H),6.92(dd,J = 8.5,2.0Hz,1H),3.79(s,3H),2.39(s,3H),2.08(s,3H)。高分辨质谱(ESI-TOF,m/z)分析结果:C18H18N2O2F3([M + H]+)的计算值为351.1320;实测值为351.1311。
