以二苯并噻吩为原料合成4-碘二苯并噻吩的一般步骤：向搅拌的、冷却至0℃的2,2,6,6-四甲基哌啶（0.25 mL，1.5 mmol）的四氢呋喃（THF，2-3 mL）溶液中依次加入1.6 M丁基锂（BuLi）的己烷溶液（1.5 mmol），5分钟后加入ZnCl2·TMEDA（0.13 g，0.50 mmol）。将反应混合物在0℃下继续搅拌15分钟，随后加入二苯并噻吩（1a，0.20 g，1.0 mmol）。在该温度下反应2小时后，加入碘（I2，0.38 g，1.5 mmol）的THF（4 mL）溶液。将混合物搅拌过夜，然后依次加入饱和碳酸氢钠（NaHCO3）水溶液和硫代硫酸钠（Na2S2O3，4 mL）溶液。用乙酸乙酯（EtOAc，3×20 mL）萃取混合物，合并有机层，用硫酸镁（MgSO4）干燥，过滤，减压浓缩。最后通过硅胶柱色谱法（洗脱剂：庚烷/二氯甲烷（CH2Cl2）梯度从100:0至80:20）纯化，得到目标产物4-碘二苯并噻吩。
