在氮气保护下,将4-溴-[2,2'-联吡啶] 1-氧化物(786 mg,3.13 mmol)溶解于干燥的氯仿(10 mL)中,并将溶液冷却至0℃。随后,向溶液中缓慢加入三溴化磷(PBr3,1.0 mL,10 mmol)。将反应混合物加热回流5小时。反应完成后,将混合物倒入冰水中,用6N氢氧化钠水溶液调节pH至碱性,并用氯仿(CHCl3)进行萃取。合并有机层,用无水硫酸钠(Na2SO4)干燥,减压浓缩得到白色固体产物4-溴-2,2'-联吡啶(728 mg,3.09 mmol,产率98.7%)。产物的结构通过1H NMR、IR光谱、熔点和ESI-MS进行了确证。1H NMR (CDCl3): δ = 8.69 (d, J = 5.0 Hz, 1H), 8.63 (d, J = 1.8 Hz, 1H), 8.49 (d, J = 5.1 Hz, 1H), 8.39 (d, J = 8.0 Hz, 1H), 7.83 (dt, J1 = 8.0 Hz, J2 = 1.6 Hz, 1H), 7.48 (dd, J1 = 5.1 Hz, J2 = 1.8 Hz, 1H), 7.34 (ddd, J1 = 7.5 Hz, J2 = 4.8 Hz, J3 = 1.6 Hz, 1H)。IR (KBr): ν = 1565, 1545, 1452, 1386, 1280, 1066, 994, 833, 787, 687 cm-1。熔点:51.5-53℃。ESI-MS: m/z = 256.9, 258.9 [M + Na]+ (计算值C10H7N2Br + Na = 256.9, 258.9)。
