以N,N'-双(2-甲基苯基)丙二酰胺(I)(0.56 g,2.0 mmol)为原料,参照文献[14]所述方法合成4-羟基-8-甲基喹啉-2(1H)-酮(II)。具体步骤如下:将化合物I溶于含有10%五氧化二磷的3.5 mL甲磺酸中,于150℃加热反应90分钟。反应完成后,使深色粘稠溶液冷却至室温。向反应混合物中加入水,长时间静置后,胶状物逐渐固化。过滤收集固体产物,将其溶于100 mL 10%氢氧化钠溶液中。过滤氢氧化钠溶液以除去不溶性杂质,随后用10%盐酸缓慢酸化至pH 4。收集析出的粗产物,用水洗涤并干燥,得到8-甲基-4-羟基喹啉-2(1H)-酮(II),为乳白色晶体(0.30 g,收率87%),熔点为357-358℃(文献值360℃[13])。产物经FT-IR(KBr,cm^-1)表征:v 3470(OH),3100(NH),1660(C=O);1H NMR(500 MHz,DMSO-d6):δ 11.25(OH),10.27(NH),7.62(d,1H,J=7.8 Hz),7.30(d,1H,J=7.2 Hz),7.01(dd,1H,J=7.8,7.1 Hz),5.27(s,1H),2.35(s,3H);13C NMR(125 MHz,DMSO-d6):δ 160.75(C=O),158.24(C-OH),139.14(C),134.10(C-CH3),130.78(CH),126.20(C-H),124.90(CH),116.17(C),99.10(CH),17.01(CH3)。元素分析(C10H9NO2)计算值:C,68.56;H,5.18;N,8.00;实测值:C,68.54;H,5.12;N,7.78。
