在0℃下,将100 mL氢氟化吡啶(Py·(HF)x,70%氟化氢和30%吡啶,购自Fluka)冷却。依次向其中加入3.7 g 2-氨基-3-羟基吡啶(分子量:110.12,33.6 mmol)和3 g NaNO2(分子量:69.00,43.5 mmol),操作需谨慎。将反应混合物在0℃下搅拌1小时,随后用10 N NaOH水溶液缓慢碱化。将混合物转移至分液漏斗中,用EtOAc萃取。合并有机相,依次用水和盐水洗涤,用Na2SO4干燥,浓缩至干。残余物通过硅胶柱色谱纯化(洗脱剂:庚烷/EtOAc,50/50),得到2.5 g(产率65%)固体2-氟-3-羟基吡啶,无需进一步纯化。薄层色谱(TLC)Rf值(EtOAc/庚烷,80/20):0.65。熔点:131℃。1H NMR(DMSO-d6,298K):δ 10.41(s,1H),7.64(td,J=1.7和4.7 Hz,1H),7.42(dd,J=1.7,1.7和10.8 Hz,1H),7.17(ddd,J=1.3,4.7和7.8 Hz,1H)。13C NMR(DMSO-d6,298K):δ 152.8(d,J1F-C=233 Hz,C),140.2(d,J2F-C=27 Hz,C),135.6(d,J3F-C=13 Hz,CH),126.2(d,J3F-C=5 Hz,CH),122.6(CH)。质谱(DCI/NH4+):C5H4FNO:131 [M+NH4+],114 [M+H+]。元素分析(C5H4FNO)符合C、H、N的理论值。
