以乙醇和2,2'-联吡啶-5,5'-二羧酸为原料合成[2,2'-联吡啶]-5,5'-二甲酸二乙酯的一般步骤:将2,2'-联吡啶-5,5'-二羧酸(200 mg,0.82 mmol)和无水乙醇(13 mL)加入干燥的烧瓶中,冰浴下搅拌。缓慢滴加亚硫酰氯(1.3 mL),随后安装回流冷凝器,加热至回流状态。反应24小时后,将反应混合物冰浴冷却,逐滴加入饱和碳酸钠溶液(20 mL)以淬灭反应。水相用二氯甲烷(4×20 mL)萃取,合并有机相并用无水硫酸钠干燥,减压浓缩。粗产物通过硅胶柱色谱法纯化(洗脱剂:3%丙酮的1:1二氯甲烷/己烷混合溶剂),得到[2,2'-联吡啶]-5,5'-二甲酸二乙酯(190 mg,产率77%)为白色固体。1H NMR (500 MHz, CDCl3) δ 9.32 (dd, J = 0.5, 2.0 Hz, 1H), 8.59 (dd, J = 0.5, 8.5 Hz, 1H), 8.46 (dd, J = 2.0, 8.5 Hz, 1H), 4.47 (q, J = 7.5 Hz, 2H), 1.46 (t, J = 7.5 Hz, 3H); 13C NMR (125 MHz, CDCl3) δ 165.2, 158.3, 150.6, 138.1, 126.6, 121.3, 61.6, 14.3; HRMS (ESI) m/z 301.1193 [C16H17N2O4 (M + H)+ 计算值301.1183]。
