将1-叔丁氧羰基哌啶-4-甲醛(35.0g,164.1mmol)溶解于二氯甲烷(200mL)中,并将溶液冷却至0℃。在搅拌下,向溶液中加入叔丁醇钾(23.9g,213mmol),随后缓慢加入碘甲烷(69.9g,492mmol)。反应混合物在0℃下搅拌30分钟后,逐渐升温至室温并继续搅拌1.5小时。反应完成后,将混合物倒入盐水(400mL)中,分离有机层。有机层用无水硫酸钠干燥,过滤后浓缩。粗产物通过硅胶柱层析纯化,得到1-Boc-4-甲酰基-4-甲基哌啶(16.36g,72.0mmol,收率43.8%)。
