以N-Boc-4-甲基-4-哌啶甲酸乙酯为原料合成1-(叔丁氧羰基)-4-甲基哌啶-4-羧酸的一般步骤:将N-Boc-4-甲基-4-哌啶甲酸乙酯(4.10 g,15.1 mmol)和氢氧化锂(2.17 g,90.6 mmol)在甲醇(30 mL)和水(20 mL)的混合溶剂中,于25℃下搅拌反应16小时。反应完成后,将反应混合物蒸发至干。将残余物在二氯甲烷和盐水之间进行分配。分离有机层,用无水硫酸镁干燥,过滤后减压浓缩,得到1-(叔丁氧羰基)-4-甲基哌啶-4-羧酸(3.38 g,产率92.0%)为白色固体。通过1H NMR和LC/MS分析确认产物结构。1H NMR (400 MHz, CDCl3): δ 3.60-3.70 (2H, m), 3.03-3.09 (2H, m), 2.05-2.08 (2H, m), 1.45 (9H, s), 1.36-1.45 (2H, m), 1.27 (3H, m); LC/MS (100%, tr = 2.500 min), m/z = 266.1 [M + Na]+ (Calc: 243)。
