以2-苯基氨基亚甲基-丙二酸二乙酯为原料合成4-羟基喹啉-3-羧酸乙酯的一般步骤:在装有机械搅拌器的1L三颈烧瓶中,加入2-苯基氨基亚甲基-丙二酸二乙酯(26.3g,0.100mol)、多磷酸(270g)和磷酰氯(750g)。将反应混合物加热至70℃,并在该温度下搅拌4小时。反应完成后,将混合物冷却至室温,然后进行过滤。将滤得的残余物用碳酸钠(Na2CO3)水溶液处理,再次过滤,用水洗涤,最后干燥。得到4-羟基喹啉-3-羧酸乙酯,为浅棕色固体(15.2g,收率70%)。
