在0℃下,向3-甲酰基氮杂环丁烷-1-羧酸叔丁酯(1.3g,27mmol)在甲醇(70ml)中的搅拌溶液中加入(1-重氮基-2-氧代丙基)膦酸二甲酯(6g,31.2mmol),随后加入碳酸钾(14.92g,108mmol)。将反应混合物在0℃下搅拌3小时。反应完成后,用乙酸乙酯(150ml)稀释反应混合物,过滤并减压浓缩,得到粗产物。粗产物通过快速色谱法(硅胶,洗脱剂:0-100%乙酸乙酯的己烷溶液)纯化,得到3-炔基-1-氮杂环丁烷甲酸叔丁酯(4.6g,94%)。1H NMR (400MHz, DMSO-d6) δ 1.34 (s, 9H), 3.22-3.23 (m, 1H), 3.34-3.39 (m, 1H), 3.68 (t, J = 6.8Hz, 2H), 4.04 (t, J = 8.4Hz, 2H)。
