中间体20-6:5-甲基吡啶-2-羧酸甲酯的合成步骤:向2,5-二甲基吡啶(3.00g,28.0mmol,CAS 589-93-5)溶于吡啶(15mL)的溶液中加入二氧化硒(4.66g,42.0mmol,CAS 7446-08-4)。将反应混合物加热回流过夜。反应完成后,冷却至室温,过滤除去固体沉淀,并依次用水和吡啶(每次5mL,共2次)洗涤。蒸发滤液,将得到的粗产物重新溶解于甲醇(100mL)中。向此溶液中加入硫酸(1.34mL,25.0mmol),然后将反应混合物加热回流5小时。反应结束后,冷却至室温,用20% NaOH水溶液碱化反应混合物。蒸发除去甲醇,加入水(50mL)稀释。用二乙醚(DEE,3×100mL)萃取混合物。合并有机相,用无水硫酸钠(Na2SO4)干燥,蒸发溶剂后,得到标题化合物5-甲基吡啶-2-羧酸甲酯(2.11g,收率50%),为浅棕色油状物。1H NMR(400MHz,CDCl3)δ 2.40(s,3H),3.97(s,3H),7.61(d,1H),8.01(d,1H),8.54(s,1H)。
