以甲醇和2-氨基-5-氟苯甲酸为原料合成2-氨基-5-氟苯甲酸甲酯的一般步骤:将2-氨基-5-氟苯甲酸(9.29 mmol,1.440 g)溶于3N HCl/MeOH溶液(30 mL)中,于100℃下加热回流过夜。反应完成后,蒸发除去溶剂,将粗产物在二氯甲烷(DCM)和饱和碳酸钾(K2CO3)水溶液之间进行萃取分离。收集有机相,蒸发浓缩后,通过硅胶(SiO2)柱色谱纯化,以己烷/乙酸乙酯混合溶剂作为洗脱剂,最终得到2-氨基-5-氟苯甲酸甲酯0.650 g,产率为42%。产物结构经1H NMR(200 MHz, CDCl3)确认:δ 3.9(s, 3H), 5.6(s, 2H), 6.6(dd, J = 9.0, 4.7 Hz, 1H), 7.0(m, 1H), 7.5(dd, J = 9.8, 3.1 Hz, 1H)。质谱分析(ESI/MS)显示分子离子峰m/z 170 [M+1]+。
