以对甲氧基苯乙酮为原料合成1-(3-溴-4-甲氧基苯基)乙酮的一般步骤:将酮7a(5.0 g,33.3 mmol)和N-溴代琥珀酰亚胺(NBS,5.9 g,33.3 mmol)溶于100 mL水中,加热至60℃。随后,缓慢滴加40% H2SO4(66.7 mmol,8.9 mL)。将反应混合物持续搅拌5小时。反应完成后,将混合物冷却至室温,用乙酸乙酯(60 mL×3)进行萃取分离。合并有机相,用饱和NaHCO3溶液洗涤,经无水MgSO4干燥后,减压浓缩,得到粗产物7b(7.5 g,收率98%),为白色固体。薄层色谱(TLC)Rf值为0.85(石油醚/乙酸乙酯,3:1)。1H NMR(400 MHz,DMSO-d6)δ:3.09(3H,s),3.93(3H,s),3.95(6H,s),7.92(1H,d,J = 8.9 Hz),7.97(1H,dd,J = 2.1 Hz,8.9 Hz),8.10(2H,d,J = 2.1 Hz);LC-MS m/z:230.9 [M + H]+。
