以2-氨基-6-氟苯并噻唑为原料合成2-氯-6-氟苯并噻唑的一般步骤:在圆底烧瓶中加入2-氨基-6-氟苯并噻唑(1.0017 g,5.96 mmol,Sigma-Aldrich Chemical Company, Inc.)、氯化铜(II)(1.201 g,8.93 mmol,Sigma-Aldrich Chemical Company, Inc.)和乙腈(ACN)中的亚硝酸叔丁酯(1.063 mL,8.93 mmol,Acros Organics)。将反应混合物加热至65℃,并在此温度下保持2小时。反应完成后,用1N盐酸稀释反应混合物,并用乙酸乙酯(EtOAc)萃取。有机相依次用水和饱和氯化钠溶液洗涤,经无水硫酸镁(MgSO4)干燥,过滤后减压浓缩,得到目标产物2-氯-6-氟苯并噻唑(0.9249 g,4.93 mmol,收率83%)。产物结构经1H NMR(400 MHz,DMSO-d6)确认:δ 7.45(td,J = 9.10, 2.74 Hz,1H),7.97-8.07(m,2H)。
