以环己酮和溴乙酸乙酯为原料合成1-羟基环己烷乙酸乙酯的一般步骤:参照例61;(1-羟基环己基)乙酸乙酯的合成;在氮气保护下,向环己酮(9.52 g,97.0 mmol)的THF(100 mL)溶液中依次加入锌粉(7.6 g,116.4 mmol)和少量碘,随后滴加溴乙酸乙酯(11.8 mL,106.7 mmol)。将反应混合物加热回流5小时。反应完成后,在冰浴冷却下缓慢加入10%硫酸溶液(100 mL),并用乙酸乙酯萃取混合物。合并有机相,用饱和碳酸氢钠水溶液洗涤,无水硫酸镁干燥,减压浓缩,得到1-羟基环己烷乙酸乙酯(17.2 g,收率94%),为无色油状物。1H-NMR(CDCl3)δ:1.27(3H,t,J = 7.0 Hz),1.35-1.71(10H,m),2.46(2H,s),3.43(1H,s),4.17(2H,q,J = 7.4 Hz)。
