将甲醇钠（295 mg，5.45 mmol）溶解于甲醇（13.6 mL）中，随后加入2,4,5-三氯嘧啶（500 mg，2.73 mmol）。将反应混合物在室温下搅拌过夜。反应完成后，将混合物减压浓缩。所得残余物用乙醚稀释，依次用1:1的水和饱和氯化铵水溶液洗涤。有机层用硫酸镁干燥，过滤后减压浓缩。最后，通过Combiflash色谱柱（洗脱剂：0-10%二乙醚/己烷）纯化，得到280 mg（1.56 mmol，收率57%）2,5-二氯-4-甲氧基嘧啶，为白色固体。
