Sodium methanolate (295 mg, 5.45 mmol) was dissolved in methanol (13.6 mL) followed by 2,4,5-trichloropyrimidine (500 mg, 2.73 mmol). The reaction mixture was stirred at room temperature overnight. After completion of the reaction, the mixture was concentrated under reduced pressure. The resulting residue was diluted with ether and washed sequentially with 1:1 aqueous solution of water and saturated aqueous ammonium chloride. The organic layer was dried with magnesium sulfate, filtered and concentrated under reduced pressure. Finally, purification by Combiflash column (eluent: 0-10% diethyl ether/hexane) gave 280 mg (1.56 mmol, 57% yield) of 2,5-dichloro-4-methoxypyrimidine as a white solid. | [References]
