以5-(苄氧基)-2-(羟甲基)-4H-吡喃-4-酮(53.2 g,229 mmol)为起始原料,将其悬浮于工业甲基化酒精(IMS,75 mL)和水(400 mL)的混合溶液中。在70℃下,向该悬浮液中缓慢加入NH4OH溶液(400 mL),并持续搅拌反应18小时。反应完成后,将反应液冷却至室温,随后用水(400 mL)稀释,并继续搅拌30分钟。通过过滤收集生成的固体产物,并在真空条件下干燥,得到目标化合物5-(苄氧基)-2-(羟甲基)吡啶-4(1H)-酮(10),为浅棕色固体(42.5 g,收率80%)。产物经LC-MS分析显示(M-H)+ = 232。1H NMR(300 MHz, DMSO-d6)数据如下:δ 11.08(br s, 1H), 7.51-7.28(m, 6H), 6.16(br s, 1H), 5.56(br s, 1H), 5.01(s, 2H), 4.34(s, 2H)。
