5-(Benzyloxy)-2-(hydroxymethyl)-4H-pyran-4-one (53.2 g, 229 mmol) was used as starting material and suspended in a mixed solution of industrial methylated alcohol (IMS, 75 mL) and water (400 mL). To this suspension, NH4OH solution (400 mL) was slowly added at 70 °C and the reaction was continuously stirred for 18 hours. Upon completion of the reaction, the reaction solution was cooled to room temperature and subsequently diluted with water (400 mL) and stirring was continued for 30 minutes. The resulting solid product was collected by filtration and dried under vacuum to afford the target compound 5-(benzyloxy)-2-(hydroxymethyl)pyridin-4(1H)-one (10) as a light brown solid (42.5 g, 80% yield). The product was analyzed by LC-MS showing (M-H)+ = 232. 1H NMR (300 MHz, DMSO-d6) data were as follows: δ 11.08 (br s, 1H), 7.51-7.28 (m, 6H), 6.16 (br s, 1H), 5.56 (br s, 1H), 5.01 (s, 2H), 4.34 (s, 2H). | [References]
