在搅拌条件下,将2,3-二氟苯甲醛(XVIII)(75.0 g,528 mmol,1.0当量)溶解于浓硫酸(565 mL)中,加热至60℃。随后,分批加入N-溴代琥珀酰亚胺(NBS)(113 g,633 mmol,1.2当量)。反应混合物在60℃下持续搅拌12小时,通过LC/MS监测确认反应完成。将反应混合物缓慢倒入冰水(500 mL)和石油醚(500 mL)的混合液中,搅拌10分钟后分离有机层。有机层在减压下浓缩,得到粗产物。粗产物通过硅胶柱色谱法(洗脱剂:100%石油醚)纯化,得到5-溴-2,3-二氟苯甲醛(XIX)(120 g,543.0 mmol,定量收率)。产物经电喷雾质谱(ESIMS)分析,确认分子式为C7H3BrF2O,m/z 221.1([M+H]+)。
