以4-羟基-2-苯基喹唑啉为原料合成4-氯-2-苯基喹唑啉的一般步骤:将4-羟基-2-苯基喹唑啉(10 mmol)与磷酰氯(10 mL)混合,在回流条件下搅拌反应9小时。反应完成后,通过减压蒸馏除去溶剂。向残余物中加入冰水,用氢氧化铵中和至沉淀形成,随后过滤收集沉淀。产物4-氯-2-苯基喹唑啉(7)通过乙醇重结晶纯化,得到棕色固体,收率为68%。产物结构通过1H NMR(500 MHz, DMSO-d6)δ 8.19-8.12(m, 3H), 7.90-7.85(m, 2H), 7.67-7.62(m, 1H), 7.61-7.55(m, 3H)和13C NMR(126 MHz, DMSO-d6)δ 161.80, 153.93, 146.17, 135.15, 132.29, 131.09, 128.78, 128.50, 127.31, 126.21, 125.70, 120.75进行确认。
