(RS)-1-[3-甲氨基-1-(2-噻吩基)]丙基]-2-萘酚性质、用途与生产工艺
(RS)-1-[3-甲氨基-1-(2-噻吩基)]丙基]-2-萘酚是盐酸度洛西汀的杂质E。盐酸度洛西汀是EliLilly公司开发的一种5-羟色胺和去甲肾上腺素再摄取抑制剂。5-羟色胺和去甲肾上腺素均属中枢神经递质,在调控情感和对疼痛的敏度方面起着重要作用。
(1)室温下向100mL的三口烧瓶中加入度洛西汀10g、乙酸乙酯50mL及浓度为12.6%的乙酸乙酯的氯化氢溶液8mL,搅拌10min,升温至35℃并保温24h,然后降温至20℃,并在该温度下过滤得到滤饼M及滤液N;
(2)将步骤(1)所得滤液N于40℃、真空度P=-0.099MPa下去减压蒸馏至无馏分馏出,得到残液F4.70g,将该残液F加入30mL乙酸乙酯中,于25℃下搅拌析晶24h,过滤得到滤饼X约4.2g及滤液Y,将滤饼X加入20mL甲醇中于20℃下搅拌析晶12h,得到滤饼S约3.5g及滤液T;
(3)将步骤(2)中的滤液T于40℃、真空度P=-0.099MPa下去减压蒸馏至无馏分馏出,得到残液W,将该残液W加入20mL乙酸乙酯中,在搅拌作用下降温至0℃析晶12h、过滤,所得滤液于50℃下去减压蒸馏,将残留的乙酸乙酯蒸除至尽,得残液约2g,即为(RS)-1-[3-甲氨基-1-(2-噻吩基)]丙基]-2-萘酚。
