Synthese
In einem Rundkolben werden 250 ml Propionsäure (Siedepunkt 141 ℃) bis zum Sieden erhitzt (Rückfluss!). Dann werden 8.5 ml Benzaldehyd hinzugegeben, und anschließend 8 ml Pyrrol unter Rühren langsam hinzugetropft. Nach der Zugabe des Pyrrols wird das Reaktionsgemisch noch für 30 Minuten unter Rückfluss gekocht, und dann auf Raumtemperatur abkühlen gelassen. Der Kolben wird anschließend im Eisbad gekühlt, damit das Tetraphenylporphyrin vollständig auskristallisiert, und dann wird über eine Glasfritte (Porosität 3) filtriert. Das abfiltrierte Rohprodukt wird 4 Mal mit je 125 ml heißem Wasser gewaschen, um die Propionsäure zu entfernen, und dann mit 25 ml Methanol, wodurch das Tetraphenylchlorin und Polymere entfernt werden. Durch Umkristallisieren aus Toluol kann man die Reinheit des erhaltenen Tetraphenylporphyrins noch weiter steigern. Die Ausbeute beträgt in der Regel zwischen 15 und 25%.
Zur Reaktion
Ab 1967 gelang Rothemund die Synthese diverser Porphyrine durch die Reaktion von Pyrrolen mit Aldehyden im Bombenrohr bei Temperaturen zwischen 140 und 220 ℃. Die Reaktionsdauer betrug hierbei 24 bis 48 Stunden und die Ausbeuten waren nie höher als etwa 10%. Darüber hinaus war durch die extremen Reaktionsbedingungen die Wahl der Edukte sehr begrenzt.
