| | Methyl Green Usage And Synthesis |
| Description | Methyl Green is a DNA stain used as a tracking dye in acid buffers for electrophoresis. It helps differentiate bacteria and is also a tissue stain for nucleoli and mitochondria in tissues. It is used with Bismarck Brown frequently. | | Chemical Properties | Methyl Green is a green microcrystalline or bright green powder with metallic luster. It is soluble in water and shows blue-green color. It is a basic dye, it is easy to combine with the high degree of polymerization of DNA to show green. Also known as double green SF. green crystals with a golden luster, or light green powder. Soluble in water, blue-green color. Slightly soluble in ethanol, insoluble in ether, pentanol. Methyl green exists in two forms. Ⅰ formula: molecular formula C26H33Cl2N3. molecular weight 458.49. green crystal, with golden yellow luster or light green powder. Soluble in water, slightly soluble in ethanol, insoluble in ether. Usually the complex salt of zinc chloride. Ⅱ formula: molecular formula C27H35BrClN3. molecular weight 516.98. green powder, soluble in water greenish blue. The solution is yellow at pH 0.2 and blue at pH 1.8. | | Uses | Methyl Green powder dye is used in various staining methods in microscopy. It is also used with Pyronine Y dye for one stage DNA (green) and RNA (red) staining method. It is also used as a counterstain with other reactions, such as demonstration of enzymatic activity and for mitochondrial staining according to Altmann. Methyl green solution is a suitable counterstain for chloroacetate esterase, nonspecific esterase, alkaline phosphatase, peroxidase, naphthol AS acetate esterase, and acid phosphatase. | | Preparation | chlorine methane and (C.I.Basic Violet 1, C.I. 42535) reaction | | Definition | ChEBI: Methylene green is an organic chloride salt having 3,7-bis(dimethylamino)-4-nitrophenothiazin-5-ium as the counterion. It stains nuclei green and is sometimes used as a counterstain to red or purple primary stains. It has a role as a fluorochrome and a histological dye. It contains a 3,7-bis(dimethylamino)-4-nitrophenothiazin-5-ium. | | Properties and Applications | green light blue. Soluble in water for blue light green, not soluble in ethanol and e alcohol. The strong sulfuric acid to red light yellow, yellow green for diluted. Dyes to join sodium hydroxide solution to colorless.
