DBCO-PEG4-amine Usage And Synthesis
DescriptionDBCO-PEG4-amine is a PEG linker which contains DBCO and amine moieties. The DBCO group is commonly used in copper-free Click Chemistry reactions. The amine group is reactive with carboxylic acids, activated NHS esters, carbonyls (ketone, aldehyde) etc. The hydrophilic PEG spacer increases the water solubility of the compound.
Biological ActivityDBCO-PEG4-amine is a PROTAC bridge and belongs to the PEG class. DBCO-PEG4-amine can be used to synthesize a series of PROTAC molecules. It is a cleavable ADC linker for the synthesis of antibody drug conjugates (ADCs).

PROTACs contain two different ligands connected by a linker; one is a ligand for an E3 ubiquitin ligase and the other is for the target protein. PROTACs exploit the intracellular ubiquitin-proteasome system to selectively degrade target proteins.
ADCs are comprised of an antibody to which is attached an ADC cytotoxin through an ADC linker.
